目的:优选纯阳正气胶囊的最佳提取工艺,研究提取液抑制沙门菌的作用及其初步机制。方法:以橙皮苷含量为评价指标,考虑乙醇溶液pH值、提取体积分数、超声时间,选用L9(34)正交进行试验结合半仿生提取技术,确定最佳提取工艺;采用平板扩散法、试管二倍稀释法(MIC法)和吸光度值测定法来观察纯阳正气胶囊提取液的体外抑菌效应。通过培养基中电导率变化,大分子物质检测验证纯阳正气胶囊抑菌初步机制。结果:第1次醇提pH3.0,第2次醇提pH8.0,提取时间25min,提取乙醇体积分数50%为最佳提取工艺;MIC为12.5 mg.mL-1,MBC为25.5 mg.mL-1。纯阳正气胶囊与空白组处理的培养基电导率、大分子物质相比分别增加了14.5%、87.1%。结论:纯阳正气胶囊提取工艺稳定、可行;提取液对沙门菌有抑制和杀菌作用。
OBJECTIVE: To optimize the extraction process of Chunyang Zhengqi Capsules and to study the inhibitory effect of the extract on Salmonella and its preliminary mechanism. Methods: The hesperidin content was taken as the evaluation index, and the optimum extraction process was determined by considering the pH value of ethanol solution, the volume fraction of extraction and the ultrasonic time. The optimum extraction process was determined by L9 (34) orthogonal test and the semi-bionic extraction technique. Tube double dilution method (MIC method) and absorbance value to observe the antibacterial effect of Chunyangzhengqi capsule extract in vitro. Through the change of conductivity in the culture medium, the main mechanism of antimicrobial activity of Chunyangzhengqi capsule was tested by macromolecular detection. Results: The optimum extraction conditions were the first alcohol extraction (pH3.0), the second alcoholysis (pH8.0) and the extraction time (25 min). The MIC was 12.5 mg.mL-1 and the MBC was 25.5 mg. mL-1. Pure Yang Zhengqi Capsules and blank group treated medium conductivity, macromolecules compared to an increase of 14.5%, 87.1%. Conclusion: The extraction method of Chunyangzhengqi Capsule is stable and feasible. The extract has inhibitory and bactericidal effects on Salmonella.
