目的探讨2-(3-羟基-1-丙酰氨基)2-脱氧-D-葡萄糖(COPADG)对食管癌裸鼠抗肿瘤作用。方法成功建立人食管癌Eca-109裸鼠模型后,随机分为4组:正常组、模型组(COPADG,200 mg·kg-1)、对照组(5-氟尿嘧啶,25 mg·kg-1)和实验组(COPADG 200 mg·kg-1+5-氟尿嘧啶25 mg·kg-1)。计算不同组的抑瘤率和细胞凋亡率,并评估裸鼠骨髓抑制及肝损伤情况。结果模型组、对照组和实验组抑瘤率分别为54.16%,38.78%,73.91%;与正常组和对照组相比,模型组和实验组的凋亡细胞率明显升高(P<0.01),显示COPADG治疗有减轻骨髓抑制及肝损伤作用。结论 COPADG有抑制食管癌肿瘤细胞增殖和血管形成及诱导细胞凋亡的作用。
Objective To investigate the antitumor effect of 2- (3-hydroxy-1-propanamido) -2-deoxy-D-glucose (COPADG) on esophageal carcinoma in nude mice. Methods The human esophageal cancer Eca-109 nude mice model was successfully established and divided into 4 groups at random: normal group, model group (COPADG, 200 mg · kg -1), control group (5-fluorouracil, 25 mg · kg -1) And experimental group (COPADG 200 mg · kg -1 + 5-fluorouracil 25 mg · kg -1). The tumor inhibition rate and apoptosis rate of different groups were calculated, and the myelosuppression and liver injury in nude mice were evaluated. Results Compared with the normal group and the control group, the rates of tumor cells in the model group, the control group and the experimental group were 54.16%, 38.78% and 73.91% , Showing that COPADG treatment has the effect of reducing myelosuppression and liver injury. Conclusion COPADG can inhibit esophageal cancer cell proliferation and angiogenesis and induce apoptosis.
