目的建立用HPLC测定氨咖黄敏胶囊中对乙酰氨基酚和咖啡因的含量。方法采用色谱柱:Diamonsil C18(5um250×4.6mm),以碳十八烷基键合硅胶为填充剂;流动相:磷酸盐缓冲液(0.01mol/L磷酸二氢钾溶液,用磷酸调节pH值至2.6±0.1):甲醇(68:32);检测波长为254nm,流速为1ml·min-1。实验结果线性范围分别是对乙酰氨基酚40.96~204.8ug·min-1(r=0.99990,n=5);咖啡因10.368~51.84ug·min-1(r=0.99963,n=5)。结论本法简单、快速、精确、重复性高,可作为氨咖黄敏胶囊的质量控制方法。
OBJECTIVE To establish a HPLC method for the determination of acetaminophen and caffeine in AHA Caramel Capsules. Methods The column was Diamonsil C18 (5um250 × 4.6mm) with carbon octadecyl-bonded silica as mobile phase. The mobile phase consisted of phosphate buffer (0.01mol / L potassium dihydrogen phosphate solution, pH adjusted with phosphoric acid To 2.6 ± 0.1): methanol (68:32); detection wavelength was 254 nm and flow rate was 1 ml · min-1. The linear range of the experimental results were acetaminophen 40.96 ~ 204.8ug · min-1 (r = 0.99990, n = 5); caffeine 10.368 ~ 51.84ug · min-1 (r = 0.99963, n = 5). Conclusion This method is simple, rapid, accurate and reproducible. It can be used as a quality control method for Ammo Caramel Capsules.
