目的:制备丹参酮微乳(Tan-ME),并考察其性质及大鼠在体肠吸收特性。方法:Zetapals多功能电位/粒度分析仪测定Tan-ME的粒度及其分布,原子力显徽镜观察微乳粒子形态。建立测定肠回流液中丹参酮Ⅱ_A的HPLC测定方法。结果:Tan-ME乳滴平均粒径为(32.25±6.59)nm,粒度均匀。肠吸收实验结果表明,Tan-ME能促进丹参酮在大鼠小肠的吸收,但Tan-ME吸收速率受Tan-ME水相含量影响较大。结论:Tan-ME制剂能够促进丹参酮在大鼠小肠的吸收,提高丹参酮的生物利用度。
OBJECTIVE: To prepare tanshinone microemulsion (Tan-ME) and to investigate its properties and intestinal absorption characteristics in rats. Methods: Zetapals multi-potential/particle size analyzer was used to determine the particle size and distribution of Tan-ME. Atomic force microscopy was used to observe the morphology of microemulsion particles. A HPLC method for the determination of tanshinone II_A in intestinal reflux was established. Results: The average particle size of Tan-ME milk droplets was (32.25±6.59) nm and the particle size was uniform. The results of intestinal absorption experiments showed that Tan-ME can promote the absorption of tanshinone in rat small intestine, but the absorption rate of Tan-ME is greatly affected by the content of Tan-ME water phase. Conclusion: Tan-ME preparation can promote the absorption of tanshinone in rat small intestine and increase the bioavailability of tanshinone.
