目的 探讨中药复方益气解毒颗粒抑制鼻咽癌细胞裸鼠移植瘤生长的分子机制。 方法 将鼻咽癌细胞系HNE1接种至BABL/c裸小鼠 ,待移植瘤生长至一定大小 (约 1cm× 1cm× 1cm) ,灌喂益气解毒颗粒药液 (按人口服药量换算鼠的给药量 ) ,一个疗程 ( 3 0d)后处死裸鼠 ,解剖获取肿瘤组织 ,测量瘤体体积大小 ,称量瘤体重量。取 10 0mg瘤体组织 ,提取总RNA后逆转录标记成cDNA探针 ,与 5 88个基因的AtlasTMHumanCancercDNAArray杂交 ,灰度扫描杂交信号及统计分析差异。 结果 益气解毒颗粒药液能明显抑制鼻咽癌细胞裸鼠移植瘤的生长 ,灌喂中药组瘤体积和重量分别为 ( 0 .2 0 7± 0 .0 2 3 )cm3 和 ( 0 .13 2± 0 .0 2 1)g ,对照组为 ( 1.3 42± 0 .2 98)cm3 和 ( 1.0 17± 0 .0 15 )g(P <0 .0 5 ) ,抑制率为 84.5 8%。HumanCancercDNAArray分析发现 ,益气解毒颗粒组有STK1、MAPK1、CDK1等 19个与细胞增殖、细胞周期、核转录因子、DNA修复相关基因表达下调。有Caspase、TNFR -1、FADD等与细胞凋亡、角化蛋白死亡受体相关基因表达上调 ,部分基因表达与未用药组组织基因表达无明显差异。 结论 益气解毒颗粒抑制鼻咽癌裸鼠移植瘤的形成 ,可能是通过抑制细胞内重要的核转录因子、细胞分裂增殖、细胞周期、DNA修复和?
Objective To investigate the molecular mechanism of Yiqi Jiedu Granule in inhibiting the growth of nasopharyngeal carcinoma xenografts in nude mice. Methods The nasopharyngeal carcinoma cell line HNE1 was inoculated into BABL / c nude mice. When the transplanted tumor grew to a certain size (about 1 cm × 1 cm × 1 cm), the nasopharyngeal carcinoma cell line HNE1 was inoculated with the solution of Yiqi Jiedu Granule Dose). After a course of treatment (30 days), the nude mice were sacrificed and the tumor tissues were dissected to obtain the tumor volume. The tumor volume was measured and the tumor weight was weighed. The total RNA was extracted from 100 mg of tumor tissue and reversely transcribed into cDNA probes. The total RNA was hybridized with Atlas TM HumanrcDNAArray of 5 88 genes, and the differences were analyzed by gray-scale scanning hybridization. Results Yiqi Jiedu Granule liquid could obviously inhibit the growth of nasopharyngeal carcinoma xenografts in nude mice. The tumor volume and weight of the Chinese herbal medicine group were (0.27 ± 0.23) cm3 and (0.13 2 ± 0. 0 2 1) g, the control group was (1.3 42 ± 0. 2 98) cm 3 and (1.0 17 ± 0 0.15) g (P 0 05), the inhibition rate was 84.5 8%. HumanCancercDNAArray analysis showed that 19 of STK1, MAPK1 and CDK1 in Yiqi Jiedu Granules group were down-regulated in the cell proliferation, cell cycle, nuclear transcription factor and DNA repair related genes. Caspase, TNFR-1, FADD and other apoptosis and keratinocyte death receptor-related gene expression was increased, some gene expression and no treatment group tissue gene expression no significant difference. Conclusion Yiqi Jiedu Granule can inhibit the formation of xenograft NPC in nude mice by inhibiting the expression of important nuclear transcription factors, cell division and proliferation, cell cycle, DNA repair and apoptosis in nasopharyngeal carcinoma.
