目的:采用高效液相色谱法建立芬格欣中核苷酸含量测定方法。方法:Diamonsil-C18柱(4.6mm×250mm,5μm),以pH4.5磷酸盐缓冲液-甲醇(96∶4)为流动相;检测波长254nm;流速1.0ml/min;柱温30℃;进样量10μl。结果:五种核苷酸与完全分离。在10.0~200.0μg/ml范围内峰面积与浓度呈良好的线性关系。精密度(RSD=0.13%)良好。平均回收率为94.6%。结论:本方法简便,迅速,准确,专属性强。
OBJECTIVE: To establish a method for the determination of nucleotide content in Fengexin by high performance liquid chromatography. METHODS: Diamonsil-C18 column (4.6 mm × 250 mm, 5 μm) was used, and the mobile phase was pH 4.5 phosphate buffer-methanol (96: 4). The detection wavelength was 254 nm. The flow rate was 1.0 ml / min. Sample volume 10μl. Results: Five nucleotides were completely separated from each other. In the range of 10.0 ~ 200.0μg / ml, there is a good linear relationship between peak area and concentration. Precision (RSD = 0.13%) is good. The average recovery was 94.6%. Conclusion: This method is simple, rapid, accurate and specific.
