目的 :研究p5 1基因的两个转录本p5 1A和p5 1B在鼻咽癌及对照组织中的表达谱 ,为进一步探讨p5 1在鼻咽癌癌变过程中的作用打下初步基础。方法 :(1)用PCR -SSCP银染技术检测 2 1例鼻咽癌组织及配对外周血标本p5 3基因的突变。 (2 )用RT -PCR检测p5 1A和p5 1B在鼻咽癌及对照组织中的表达。结果 :(1) 2 1例鼻咽癌组织及配对外周血标本p5 3基因未发现突变。 (2 )在鼻咽癌及对照组织中 ,p5 1B的mRNA表达要高于p5 1A。 (3)p5 1A和p5 1BmRNA在鼻咽癌组织中的表达高于对照组织。结论 :(1)p5 1A和p5 1BmRNA在鼻咽癌中的表达高于对照组织。 (2 )p5 1A和p5 1BmRNA表达的改变与p5 3基因的突变无关
OBJECTIVE: To study the expression profiles of two transcripts p5 1A and p5 1B in nasopharyngeal carcinoma and control tissues, and lay a preliminary foundation for further exploring the role of p5 1 in the carcinogenesis of nasopharyngeal carcinoma. Methods: (1) The mutation of p5 3 gene in 21 cases of nasopharyngeal carcinoma and paired peripheral blood samples was detected by PCR-SSCP silver staining. (2) The expression of p5 1A and p5 1B in nasopharyngeal carcinoma and control tissues was detected by RT-PCR. Results (1) No mutation was found in p1 gene of 21 cases of nasopharyngeal carcinoma and paired peripheral blood samples. (2) In NPC and control tissues, mRNA expression of p5 1B was higher than that of p5 1A. (3) The expression of p5 1A and p5 1B mRNA in NPC tissues was higher than that in control tissues. Conclusion: (1) The expression of p5 1A and p5 1B mRNA in NPC is higher than that in control. (2) The changes of p5 1A and p5 1B mRNA expression have nothing to do with the mutation of p5 3 gene
