以山桐子(Idesia polycarpa)的幼根为外植体,采用改良的MS培养基(除钙盐外,其余大量元素用MS的2/3量)附加不同浓度激素组合研究山桐子离体培养技术。结果表明,以改良的MS+TDZ0.02mg·L-1(单位下同)+NAA0.5+6-BA0.1诱导愈伤组织为最佳;以改良的MS+TDZ0.05+NAA0.1+6-BA1.0进行芽的分化与增殖为最佳;以改良的MS+TDZ0.02+NAA0.2+6-BA1.0进行壮苗培养为最佳;在1/2MS+NAA0.8+IBA0.3培养基上生根,最终再生成完整植株。
Using the roots of Idesia polycarpa as explants, we studied the effects of different concentrations of hormones on the in vitro culture of Jatropha curcas by using modified MS medium (except calcium salts, other 2/3 of MS) . The results showed that the best MS + TDZ0.02mg · L-1 (the same unit below) + NAA0.5 + 6-BA0.1 induced callus the best; with improved MS + TDZ0.05 + NAA0.1 + 6-BA1.0 is the best bud differentiation and proliferation; best MS + TDZ0.02 + NAA0.2 + 6-BA1.0 for seedling culture is the best; in 1 / 2MS + NAA0.8 + IBA0.3 medium rooting, and ultimately regenerate the entire plant.
