目的:观察补肝方对亚健康大鼠模型下丘脑单胺类神经递质含量及海马脑区谷氨酸受体(NR2A mRNA)表达的影响。方法:采用游泳运动、睡眠剥夺和夹尾刺激等复合因素制造亚健康大鼠模型,分别以生理盐水、补肝方高、低剂量(含生药20,10 g.kg-1)ig,1次/d,连续20 d,检测下丘脑多巴胺(DA),5-羟色胺(5-HT)含量及海马脑区NR2A mRNA表达。结果:模型组下丘脑DA含量明显低于正常组(P<0.01),补肝方高、低剂量组下丘脑DA含量均高于模型组(P<0.01)。模型组下丘脑5-HT含量较正常组明显增高(P<0.01);补肝方高、低剂量组比模型组有明显降低(P<0.01)。模型组NR2A mRNA表达明显低于对照组(P<0.01),补肝方有升高NR2A mRNA的趋势。结论:补肝法改善疲劳、烦躁、失眠等表现的机制可能与调节DA和5-HT的异常,上调海马NR2A mRNA的表达,抑制兴奋性氨基酸类神经递质的传导异常有关。
Objective: To observe the effect of Bugan Fang on monoamine neurotransmitter in hypothalamus and expression of glutamate receptors (NR2A mRNA) in hippocampus of sub-healthy rats. Methods: Sub-healthy rats were made by swimming, sleep deprivation and tail-tailed stimulation. The rats were randomly divided into three groups: normal saline, Buganfang high and low dose (including crude drug 20,10 g.kg-1) / d for 20 days. The contents of dopamine (DA), serotonin (5-HT) and NR2A mRNA in the hippocampus were detected. Results: The content of DA in hypothalamus in model group was significantly lower than that in normal group (P <0.01). The content of DA in hypothalamus of high and low dose of Bugan Fang was higher than that in model group (P <0.01). The content of 5-HT in the hypothalamus of the model group was significantly higher than that of the normal group (P <0.01). The levels of 5-HT in the hypothalamus of the model group were significantly lower than those in the model group (P <0.01). NR2A mRNA expression in the model group was significantly lower than that in the control group (P <0.01), Bugan Decoction increased NR2A mRNA tendency. Conclusion: The mechanism of Bugan method for improving fatigue, irritability and insomnia may be related to the abnormalities of DA and 5-HT, the upregulation of NR2A mRNA in hippocampus and the abnormal conduction of excitatory amino acid neurotransmitters.
