目的探讨claudin-7及MMP-9在结肠癌组织中的表达及其与临床病理参数之间的关系。方法应用免疫组化s-p法检测claudin-7及MMP-9在40例结肠癌患者、20例结肠腺瘤患者及10例正常人的结肠组织中的表达。结果正常结肠黏膜、结肠腺瘤及结肠癌组织中claudin-7阳性表达率呈递减趋势,组间差异有统计学意义(χ2=9.214,P<0.05);MMP-9阳性表达率呈递增趋势,组间差异有统计学意义(χ2=16.406,P<0.01)。claudin-7及MMP-9的表达均与肿瘤的分化程度、浸润深度及有无淋巴结转移有关(P<0.05),而在年龄、性别、肿瘤大小及发生部位组内差异均无统计学意义(P>0.05)。Spearman相关分析显示结肠癌组织中claudin-7与MMP-9蛋白表达呈负相关(χ2=-0.453,P<0.01)。结论 claudin-7的低表达和MMP-9的过表达在结肠癌的发生、发展及其浸润和转移中具有重要作用。
Objective To investigate the expression of claudin-7 and MMP-9 in colon carcinoma and its relationship with clinicopathological parameters. Methods The expressions of claudin-7 and MMP-9 were detected by immunohistochemical s-p method in 40 colon cancer patients, 20 colon adenoma patients and 10 normal human colon tissues. Results The positive expression rate of claudin-7 in normal colorectal mucosa, colon adenoma and colon cancer showed a decreasing trend with statistical significance (χ2 = 9.214, P <0.05). The positive expression rate of MMP-9 showed an increasing trend, The difference between the two groups was statistically significant (χ2 = 16.406, P <0.01). The expression of claudin-7 and MMP-9 were correlated with tumor differentiation, depth of invasion and lymph node metastasis (P <0.05), but there was no significant difference in age, sex, tumor size and site of occurrence P> 0.05). Spearman correlation analysis showed that the expression of claudin-7 and MMP-9 in colon cancer tissues was negatively correlated (χ2 = -0.453, P <0.01). Conclusion The low expression of claudin-7 and the overexpression of MMP-9 play an important role in the carcinogenesis, development, infiltration and metastasis of colon cancer.
