目的:研究扇贝裙边糖胺聚糖(SS-GAG)抗动脉粥样硬化作用机制。方法:将培养的U937细胞分为正常组、模型组和SS-GAG不同浓度组,除正常组外,其余组加入氧化低密度脂蛋白造模后给予相应浓度药物进行处理,分别采用不同方法测定各组细胞超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性和一氧化氮(NO)含量及Ca2+水平。结果:各组中SOD含量无显著性差别;SS-GAG组中GSH-Px活性和NO含量与模型组比较明显升高,而细胞内Ca2+水平则明显降低。结论:SS-GAG可能通过增强GSH-Px活性和NO释放,同时降低细胞内Ca2+水平而发挥抗动脉粥样硬化作用。
Objective: To study the antiatherosclerotic mechanism of scallop skirt glycosaminoglycans (SS-GAG). Methods: The cultured U937 cells were divided into normal group, model group and different concentrations of SS-GAG group. Except the normal group, the other groups were treated with oxidized low-density lipoprotein (LDL) and then given the corresponding concentrations of drugs for treatment. The activities of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and nitric oxide (NO), and Ca2+ levels in various groups of cells. Results: There was no significant difference in the content of SOD in each group. The activity of GSH-Px and the content of NO in SS-GAG group were significantly higher than those in model group, but the level of intracellular Ca2+ was significantly decreased. Conclusion: SS-GAG may play anti-atherosclerotic effects by enhancing the activity of GSH-Px and NO, and reducing the intracellular Ca2+ level.
