目的:探讨不同热缺血时间下供心冷保存期间ATP的下降程度,以确定不同热缺血时间下供心冷保存的时限。方法:新西兰大白兔18 只,随机分成1,2,3 组(n= 6) ,开胸后,取左室前壁心肌测ATP,按常规取心方法分别在阻断主动脉后( 热缺血)0 min(1 组) ,5 min(2 组) ,10 min(3 组) 取下心脏,保存于4℃的Ringer’s 液中,30 min,1 h,2 h,3 h,4 h 取左室心肌测ATP( 其中1 组加测5 h) 。结果:热缺血0 min,心脏保存于4 ℃Ringer’s 液中,ATP下降至缺血前50% 约为4~5 h,热缺血5 min,约为3 h,热缺血10 min,约为1 h。结论:用4℃的Ringer’s 液保存供心,热缺血0 min,冷存时限为3 ~4 h;热缺血5 min,冷存时限为2 h;热缺血应小于10 min。
OBJECTIVE: To investigate the decrease of ATP during cardiomyocyte storage under different warm ischemia to determine the time frame for cold preservation under different warm ischemia. Methods: Eighteen New Zealand white rabbits were randomly divided into 1, 2 and 3 groups (n = 6). After thoracotomy, the myocardium of the anterior chamber of the left ventricle was measured for ATP. After aortic occlusion Blood samples were collected from Ringer’s solution at 4 ° C for 30 min, 1 h, 2 h, 3 h, 4 h h to take left ventricular myocardial ATP (including 1 group plus test 5 h). Results: During warm ischemia, the hearts were stored in Ringer’s solution at 4 ℃. ATP decreased to about 50% before ischemia, about 4 ~ 5 hours, warm ischemia for 5 minutes, about 3 hours, and warm ischemia for 10 minutes , About 1 h. CONCLUSION: The donor hearts were preserved with Ringer’s solution at 4 ° C for 0 min and the cold storage time was 3-4 h. The warm ischemia was 5 min and the cold storage time was 2 h. The warm ischemia should be less than 10 min.
