目的:探讨高危型HPV16 E6蛋白的表达对宫颈癌细胞株中p63α表达的影响。方法:用RT-PCR法从SiHa中扩增出带酶切位点的HPV16 E6的片段,将HPV16 E6基因片段克隆到p3×flag-CMV-Myc-24真核表达载体上,通过脂质体转染入宫颈癌细胞株后,W estern blot检测p63α的表达。结果:成功构建HPV16 E6真核表达载体,转染至宫颈癌SiHa和Caski细胞株中,p63α均低于对照组。结论:高危型HPV E6能够抑制p63α的表达,提示p63α与HPV E6的相互作用在子宫颈癌发病中发挥重要作用。
Objective: To investigate the effect of high-risk HPV16 E6 protein expression on the expression of p63α in cervical cancer cell lines. Methods: The fragment of HPV16 E6 with restriction enzyme sites was amplified by RT-PCR from SiHa. The HPV16 E6 gene fragment was cloned into p3 × flag-CMV-Myc-24 eukaryotic expression vector, After transfection into cervical cancer cell lines, the expression of p63α was detected by Western blot. Results: The eukaryotic expression vector of HPV16 E6 was successfully constructed and transfected into SiHa and Caski cell lines, both of which were lower than the control group. Conclusion: High-risk HPV E6 can inhibit the expression of p63α, suggesting that the interaction between p63α and HPV E6 plays an important role in the pathogenesis of cervical cancer.
