目的检测胃癌组织中p15基因及启动子甲基化状态和P15蛋白表达情况。方法选择p15基因及启动子区域,用PCR-SSCP,MSP(甲基化特异的PCR)法、测序和免疫组化等方法对100例胃癌患者的癌组织和癌旁组织进行检测。结果 11%的病例P15表达阴性,9%的病例具有p15基因启动子区的高甲基化,9%的病例同时有P15表达阴性和p15基因启动子区的高甲基化,p15异常与低分化胃癌有关,p15基因发现两例DNA序列改变。结论 p15基因在胃癌中起一定作用且p15基因启动子区域高甲基化是胃癌中p15基因失活的主要原因。
Objective To detect the p15 gene and promoter methylation status and P15 protein expression in gastric cancer. Methods The p15 gene and its promoter region were selected. PCR and SSCP, MSP (methylation-specific PCR), sequencing, and immunohistochemistry were used to detect the tumor tissues and paracancerous tissues of 100 patients with gastric cancer. Results P15 was negative in 11% of the cases, 9% of the cases had hypermethylation of the promoter region of p15 gene, and 9% of cases had both negative P15 expression and hypermethylation of the promoter region of p15 gene. p15 abnormalities were associated with poorly differentiated gastric cancer. The p15 gene revealed two changes in DNA sequence. Conclusion The p15 gene plays a role in gastric cancer and hypermethylation of p15 gene promoter region is the main reason for the inactivation of p15 gene in gastric cancer.
