用生物素-亲合素酶联法(BAE)测定正常大鼠肝细胞表面HDL受体的特性。实验结果表明,无载脂蛋白E高密度脂蛋白(apoEHDL)与大鼠肝细胞具有高亲和力、高专一性结合,其平衡解离常数Kd=5.52mg/L,最大结合量B(max)=173μg/g细胞蛋白。本法灵敏度高、反应温和,不影响HDL与其受体的结合能力。
The characteristics of HDL receptors on the surface of normal rat hepatocytes were determined by biotin-avidin assay (BAE). The results showed that apoEHDL without apolipoprotein E had high affinity and high specific binding to rat hepatocytes. The equilibrium dissociation constant Kd was 5.52 mg / L and the maximum binding capacity B (max) ) = 173 μg / g cellular protein. The law of high sensitivity, mild reaction, does not affect the binding capacity of HDL and its receptors.
