盾叶薯蓣是中国特有的甾体激素类药源植物,但关于其种质资源遗传多样性和分子标记辅助育种的研究相对较少。本研究在盾叶薯蓣转录组分析的基础上,进一步挖掘其SNP和SSR分子标记。研究发现,盾叶薯蓣转录组的37 115条unigene中含有124 692个SNP位点,平均每条unigene含3.36个SNP位点。用软件MISA对盾叶薯蓣转录组进行搜索发现13 514个SSR位点分布于11 615条unigene序列中,SSR发生频率为13.66%,平均每7.18 kb含1个SSR位点。共筛选设计出4 329对SSR引物。本研究不仅丰富了盾叶薯蓣的分子标记,为其遗传资源评价、种质资源鉴定与改良提供帮助,而且对盾叶薯蓣功能基因资源的开发利用及其比较基因组学研究具有重要的价值。
Dioscorea zingiberensis is a steroidal hormone-specific plant in China, but there are relatively few studies on its genetic diversity and molecular marker-assisted breeding. In this study, based on the analysis of transcriptome of Dioscorea zingiberensis, further SNP and SSR molecular markers were excavated. It was found that there were 124 692 SNP sites in 37 115 unigene of Dioscorea zingiberensis transcriptome with an average of 3.36 SNPs per unigene. Searching the transcriptome of Dioscorea zingiberensis using software MISA found that 13 514 SSR loci were found in 11 615 unigene sequences, with an SSR frequency of 13.66% and an average of one SSR locus per 7.18 kb. A total of 4 329 pairs of SSR primers were designed and screened. This study not only enriches the molecular marker of Dioscorea zingiberensis, but also helps to evaluate its genetic resources, identification and improvement of germplasm resources, and is of great value to the development and utilization of Dioscorea zingiberensis functional gene resources and its comparative genomics.
