目的:为应用tPA单克隆抗体建立ELISA法检测tPA含量以及研究tPA功能和结构的关系。方法:运用杂交瘤技术成功地研制5株tPA单克隆抗体,并进行较系统的免疫特性的鉴定。结果:5株单抗特异性高,与uPA、PLG、Fg、Fb、BSA均无交叉反应;亲合力强1H4>3C10>5H10>4E6>4C6;腹水效价5×10-6~1×10-7;免疫球蛋白亚类为IgG1和IgG2a;5株单抗中,3C10和1H4可明显抑制tPA活性,而5H10、4E6、4C6则对tPA活性无明显影响。结论:为进一步应用这些单抗作为研究手段提供了基础。
OBJECTIVE: To establish an ELISA assay for the content of t-PA and to study the relationship between t-PA function and structure for the application of t-PA monoclonal antibody. Methods: Five monoclonal antibodies against tPA were successfully developed by hybridoma technique and identified more systematically. Results: The 5 McAbs were highly specific and had no cross reaction with uPA, PLG, Fg, Fb and BSA. The avidity was 1H4> 3C10> 5H10> 4E6> 4C6. The ascites titer was 5 × 10-6 ~ 1 × 10-7; immunoglobulin subclass IgG1 and IgG2a; 5 monoclonal antibody, 3C10 and 1H4 can significantly inhibit t-PA activity, while 5H10, 4E6, 4C6 T-PA activity had no significant effect. Conclusion: This study provides the basis for further application of these mAbs as research tools.
