Non-pathogenic Gram-positive oral commensal bacteria expressing recombinant fusion proteins on their cell surface colonize the oropharynx that elicit mucosal and systemic immune responses to exogenous antigens. In this system, the fusion protein vaccine is released and immobilized on the surface of the commensal bacteria, whereas commensal bacteria colonize the mucosal site where the specific pathogen invades. The expression of these foreign proteins is accomplished by a common mechanism by which gram-positive bacteria transfer and immobilize them on the cell surface. This provides a safe method for utilizing engineered pathogens as live vaccine release vectors.
