目的:探讨黄癸素(HB)在体外对血管生成的影响。方法:MTT法测定黄癸素对ECV304增殖的影响,划痕法检测黄癸素对ECV304细胞迁移的影响,二维培养观察黄癸素对ECV304细胞小管样结构形成的影响。结果:MTT法测得黄癸素对ECV304细胞24、48、72小时的IC50分别为40.61、7.15和2.40mg/L;划痕法测得黄癸素浓度为1.25、2.5、5mg/L时的迁移抑制率分别为39%、53%和79%;与阴性对照组比较,经黄癸素处理的ECV304细胞小管数目均显著减少,且管腔不完整,浓度为20、30、40mg/L时小管形成抑制率分别为26.99%、39.26%和65.64%。结论:黄癸素可直接抑制ECV304细胞的增殖、迁移和小管样结构的形成,抑制血管新生,从而可能对肿瘤血管的形成起到间接抑制作用。
Objective: To investigate the effect of HB on the angiogenesis in vitro. Conclusion: Decoction of Trichoderma direct inhibition of ECV304 cell proliferation, migration and tubular-like structure formation, inhibition of angiogenesis, which may play an indirect role in the formation of tumor blood vessels.
