本文作者利用[~3H]GBR12935测定新纹状体中DAUC(Dopamine Uptake Complex)的密度,探讨锌对GBR12935与DAUC的结合、DAUC对多巴根(DA)的摄取和DAUC上可卡因亲和状态的调控作用。结果表明:锌可明显增加[~3H]GBR12935与DAUC的结合;外源性锌增加DAUC上可卡因R_H的比例(由73%±4%增至90%±2%),降低K_H(由2250±880nM降至890±150nM);抑制DAUC对DA的摄取;加强可卡因对DA摄取的抑制效应。
The authors used [~ 3H] GBR12935 to determine the density of DAUC (Dopamine Uptake Complex) in new striatum and discussed the effect of zinc on the binding of GBR12935 to DAUC, the uptake of DAUC by DAUC and the affinity of cocaine on DAUC Regulation. The results showed that: zinc significantly increased the binding of [~ 3H] GBR12935 to DAUC; exogenous zinc increased the ratio of cocaine R_H on DAUC (from 73% ± 4% to 90% ± 2%) and decreased K_H (from 2250 ± 880nM down to 890 ± 150nM); inhibition of DAUC uptake of DA; enhance the inhibitory effect of cocaine on DA uptake.
