目的:研究杏仁核点燃癫痫模型中c-Jun氨基末端激酶(JNK)通路的活化。方法:雄性Wistar大鼠20只,随机分为对照组10只和模型组10只。将电极植入大鼠右侧杏仁核,对照组不给予电刺激,模型组每天给予500μA的电流刺激,大鼠连续10 d在刺激后达到5级发作视为点燃成功。成功点燃的大鼠,在最后1次5级发作后2 h处死。Western blot检测2组海马JNK、p-JNK、c-Jun、p-c-Jun的表达。结果:模型组刺激侧海马p-JNK、p-c-Jun表达明显高于对照组(P<0.05)。结论:p-JNK/p-c-Jun通路可能参与杏仁核癫痫模型点燃。
AIM: To investigate the activation of c-Jun N-terminal kinase (JNK) pathway in amygdala-kindled epilepsy model. Methods: Twenty male Wistar rats were randomly divided into control group (n = 10) and model group (n = 10). The electrodes were implanted into the right amygdala of rats. The control group received no electrical stimulation. The model group was given 500μA current stimulation every day, and the rats reached the grade 5 attack after 10 days’ continuous stimulation. Successful lit rats were sacrificed 2 h after the last level 5 attack. Western blot was used to detect the expression of JNK, p-JNK, c-Jun and p-c-Jun in the two groups. Results: The expression of p-JNK and p-c-Jun in hippocampus of the model group was significantly higher than that of the control group (P <0.05). Conclusion: The p-JNK / p-c-Jun pathway may be involved in the amygdala model of epilepsy.
