目的:观察心肌缺血再灌注后心肌细胞凋亡与一些细胞活素水平变化的关系,以及黄芪的作用。方法:69只大耳白家兔,随机分为心肌缺血-再灌注组、黄芪治疗组、心肌缺血坏死组、假手术对照组。采用脱氧核糖核苷酸末端转移酶介导的缺口末端标记法和心肌细胞DNA电泳检测心肌组织的细胞凋亡情况,并应用MTT法检测外周血单个核细胞分泌IL-1、IL-4、TNF、IFNγ的水平。结果:缺血再灌注后,凋亡的心肌细胞增加,同时伴有IL-1、TNF、IFNγ升高和IL-4水平降低。黄芪治疗后只有少量的细胞凋亡及一些细胞活素的轻度变化。结论:心肌缺血再灌注心肌细胞凋亡过程中,细胞活素可能有重要的作用;黄芪可抑制再灌注时的心肌细胞凋亡,此作用可能与这些细胞活素的水平变化有关。
OBJECTIVE: To observe the relationship between myocardial cell apoptosis and the changes of some cytokines after myocardial ischemia and reperfusion, and the role of jaundice. Methods: 69 large white rabbits were randomly divided into myocardial ischemia-reperfusion group, astragalus treatment group, myocardial ischemic necrosis group and sham-operated control group. Apoptosis of myocardial tissue was detected by nick end labeling method mediated by deoxyribonucleotide terminal transferase and DNA electrophoresis of myocardial cells, and IL-1, IL-4 and TNF secreted by peripheral blood mononuclear cells were detected by MTT assay. , IFNγ levels. RESULTS: After ischemia and reperfusion, the number of apoptotic cardiomyocytes increased, accompanied by elevated IL-1, TNF, and IFNγ and decreased IL-4 levels. After treatment with jaundice, only a small amount of apoptosis and slight changes in some cytokines were observed. Conclusion: Cytokines may play an important role in myocardial apoptosis induced by myocardial ischemia and reperfusion. Huangqi can inhibit cardiomyocyte apoptosis after reperfusion, which may be related to the changes of these cytokines.
