目的:优选纤维素酶协同超声波法提取复方半边莲注射液的工艺条件。方法:在超声波条件固定的情况下,采用正交试验考察酶用量、酶解pH、酶解时间及酶解温度对复方半边莲注射液中野黄芩苷提取率的影响。采用HPLC测定野黄芩苷含量。结果:最佳工艺条件为酶解温度40℃,酶解pH 6.0,酶用量1 g(6.7 mg·g-1),酶解时间30 min;野黄芩苷质量浓度4.68 g·L-1,明显高于煎煮法(1.34 g·L-1)、单一超声波法(3.84 g·L-1)和单一纤维素酶法(2.26 g·L-1)。结论:纤维素酶协同超声波法提取复方半边莲注射液具有效率高、生产成本低、生产周期短等优点。
OBJECTIVE: To optimize the technological conditions of extraction of Compound Lobelia Compound by cellulase and ultrasonic wave. Methods: The effects of enzyme dosage, pH, enzymolysis time and enzymolysis temperature on the extraction efficiency of baicalin from compound Chinese cabbage extract were studied by orthogonal test under the condition of fixed ultrasound. Determination of wild baicalin by HPLC. Results: The optimum conditions were enzymolysis temperature 40 ℃, pH 6.0, enzyme dosage 1 g (6.7 mg · g -1), enzymolysis time 30 min, and the concentration of wild baicalin 4.68 g · L -1 (1.34 g · L -1), single ultrasonic method (3.84 g · L -1) and single cellulase (2.26 g · L -1). Conclusion: The combination of cellulase and ultrasonic extraction of compound Chinese lobelia has the advantages of high efficiency, low production cost and short production cycle.
