目的:探究微重力对干细胞向神经元分化的影响,为临床治疗神经退行性疾病提供新的思维和方法。方法:运用流式细胞术鉴定骨髓间充质干细胞及其微重力对凋亡的影响。运用反转录PCR检测微重力干预干细胞48h及72h后神经营养因子BDNF,CNTF,NGF的表达。运用ELISA检测其分泌营养因子的浓度。结果:流式细胞仪检测细胞凋亡,两组之间并无差异。CNTF分泌在微重力干预后有明显增加(p<0.05)。BDNF浓度有轻度增加。结论:模拟微重力可以促进骨髓间充质干细胞神经营养因子的表达。
Objective: To explore the effect of microgravity on the differentiation of stem cells into neurons and to provide new thinking and methods for clinical treatment of neurodegenerative diseases. Methods: Flow cytometry was used to identify the effects of bone marrow mesenchymal stem cells and their microgravity on apoptosis. Reverse transcriptional PCR was used to detect the expression of BDNF, CNTF and NGF after 48h and 72h intervention by microgravity. The secretion of trophic factor was detected by ELISA. Results: Flow cytometry showed that there was no difference between the two groups. CNTF secretion significantly increased after microgravity intervention (p <0.05). BDNF concentration increased slightly. Conclusion: Simulated microgravity can promote the expression of neurotrophic factor of bone marrow mesenchymal stem cells.
