在以棉花基因组 DNA(gDNA)为探针的基因组原位杂交(genomic in situ hybridization, GISH)实验中, 观察到 6 个 NOR(nucleolar organizer region)信号. 在对草棉或陆地棉的同一有丝分裂细胞分别进行以 45S rDNA和 gDNA 为探针的原位杂交中, 发现以 gDNA为探针所产生的 NOR 信号与以 45S rDNA为探针所产生的 NOR 信号在数目、位置以及大小方面极其相似甚至相同, 由此将以 gDNA为探针所产生的NOR命名为GISH-NOR. 在棉花GISH-NOR中, 陆地棉和雷蒙德氏棉全部为端部类型GISH-NOR,而对于草棉变种阿非利加棉则为 4 个端部类型和 2 个着丝粒类型 GISH-NOR. 陆地棉 6 个 GISH-NOR中的 2 个位于 A 亚组染色体上, 其余的 4 个位于 D 亚组染色体上. 在以雷蒙德氏棉为靶 DNA, 以其本身 gDNA 为探针的 GISH 中, 观察到 6 个 GISH-NOR 信号. 而在以阿非利加棉为靶 DNA, 以其本身gDNA为探针的GISH中, 没有观察到GISH-NOR信号, 并且有一对染色体长臂近一半区域不显示信号.在以陆地棉为靶 DNA, 阿非利加棉为探针时发现, 如果用 D 基因组棉种作封阻, 则不出现 GISH-NOR信号; 如果改用鲑鱼精 DNA作封阻, 则出现 GISH-NOR 信号. 而在以 D 基因组二倍体棉种戴维逊氏棉为探针时, 即使用 A基因组棉种作封阻, 也能观察到 6 个
