采用水提醇沉法从沙棘叶中提取水溶性沙棘叶多糖(WPFH),通过响应面优化蒽酮-硫酸显色,用分光光度法测定沙棘叶水溶性多糖的总糖含量。通过单因素和响应面法确定了最佳蒽酮-硫酸显色条件:显色温度为100℃,蒽酮试剂的用量为3.54mL,显色时间为10.95min。测定波长620nm,葡萄糖质量浓度在6.67—160μg/mL范围内与吸光度呈良好的线性关系,r=0.9993。该方法简单可行,平均回收率为100.36%,RSD为2.75%。精制WPFH换算因子f为1.266。
Soluble Seabuckthorn Leaf Polysaccharides (WPFH) were extracted from sea buckthorn leaves by water extraction and alcohol precipitation. The anthrone - sulfuric acid color was optimized by response surface, and the total sugar content of water - soluble polysaccharides was determined by spectrophotometry. The optimal anthrone-sulfuric acid color conditions were determined by single factor and response surface method. The color temperature was 100 ℃, the anthrone reagent dosage was 3.54mL and the color development time was 10.95min. The determination of the wavelength of 620nm, glucose concentration in the range of 6.67-160μg / mL and absorbance showed a good linear relationship, r = 0.9993. The method is simple and feasible, with an average recovery of 100.36% and RSD of 2.75%. Refined WPFH conversion factor f is 1.266.
