目的研究一种双链季铵盐消毒剂的杀菌效果及作用机理。方法采用载体定量杀菌试验和扫描电镜观察的方法,对该季铵盐化合物杀灭金黄色葡萄球菌和大肠杆菌的效果及其超微结构进行了实验室观察。结果用100 mg/L该双链季铵盐消毒剂作用5 min,对载体(纸片)上金黄色葡萄球菌和大肠杆菌的杀灭率均达到100%。通过扫描电镜观察发现,经含量25 mg/L的该双链季铵盐消毒剂作用20 min的金黄色葡萄球菌菌体仍保有较完好的外部形态,但菌体表面粗糙,有少数菌体胞壁出现皱褶;可见到大肠杆菌表面出现凹陷和皱缩。经含量100 mg/L的该双链季铵盐消毒剂作用20 min的金黄色葡萄球菌的细胞壁有显著皱褶和破裂,表面有许多颗粒状物出现,形成小泡和小球;大肠杆菌菌体局部断裂破损。结论该双链季铵盐消毒剂在较低浓度具有快速杀灭细菌繁殖体的效果,其致死细菌可能与细胞质的凝集和胞内物质的渗漏有关。
Objective To study the bactericidal effect and mechanism of a double-chain quaternary ammonium disinfectant. Methods The effect of the quaternary ammonium compounds on killing Staphylococcus aureus and Escherichia coli and the ultrastructure of the quaternary ammonium compounds were observed by the method of quantitative bactericidal test and scanning electron microscopy. The double stranded quaternary ammonium salt disinfectant with a content of 100 mg / L for 20 min had significant folds and ruptures in the cell wall, and many granules appeared on the surface to form vesicles and globules. Conclusion The double-chain quaternary ammonium salt disinfectant has the effect of rapidly killing bacterial propagules at lower concentration, and the lethal bacteria may be related to the cytoplasm agglutination and the leakage of intracellular substances.
