目的:探讨补肾益气活血方-葆青康对应激、老年小鼠学习与记忆及相关酶活性的影响。方法:采用游泳实验、耐缺氧实验和耐高温实验观察葆青康对小鼠抗应激能力的提升作用;采用食饵法大迷宫法测定葆青康对中、老年小鼠学习与记忆的影响;采用McEwen法测定葆青康对中、老年小鼠脑中单胺氧化酶-B(MAO-B)活性的影响;采用TBA比色法、邻苯三酚自氧化法和DNBT法测定葆青康对老年小鼠肝丙二醛(MDA)含量,心、脑、肝和血中超氧化物歧化酶(SOD)活性和谷胱甘肽过氧化物酶(GSH-Px)活性的影响。结果:葆青康能延长小鼠游泳时间、增加常压缺氧小鼠存活时间和降低小鼠耐高温的死亡率,提高小鼠抗应激的能力;葆青康能增加中老年小鼠学习与记忆能力,降低中老年小鼠脑MAO-B活性,降低老年小鼠肝中MDA含量、提高老年小鼠心、脑、肝和血中SOD和GSH-Px活性。结论:葆青康具有抗应激和抗衰老作用。
Objective: To investigate the effect of Bushen Yiqi Huoxue Fang-Qi Qingkang on learning, memory and related enzyme activities in stress and aged mice. Methods: The effects of Qiqingkang on the anti-stress ability of mice were observed by swimming experiment, hypoxia tolerance test and high temperature resistance test; the effect of Qiqingkang on learning and memory of middle-aged and aged mice was determined by predatory method. The McEwen method was used to determine the effect of Qiqingkang on the activity of monoamine oxidase-B (MAO-B) in the brains of middle-aged and aged mice. The TBA colorimetric method, pyrogallol autoxidation method and DNBT method were used to determine the old-age Kang of old age. Malondialdehyde (MDA) content in the liver, superoxide dismutase (SOD) activity, and glutathione peroxidase (GSH-Px) activity in heart, brain, liver, and blood. Results: Qiqingkang can prolong the swimming time of mice, increase the survival time of normoxic and hypoxic mice, and reduce the mortality rate of mice with high temperature resistance, and improve the ability of mice to resist stress; Qi Qingkang can increase the learning of middle-aged and old mice. With memory ability, the activity of MAO-B in the brain of middle-aged and aged mice was reduced, the content of MDA in the liver of aged mice was reduced, and the activities of SOD and GSH-Px in heart, brain, liver and blood of aged mice were increased. Conclusion: Qiqingkang has anti-stress and anti-aging effects.
