目的 研究帕金森病 (PD)大鼠的神经行为学特点及其中脑酪氨酸羟化酶 (TH)mRNA表达的改变。 方法 利用阿朴吗啡诱发旋转试验和前肢功能试验 ,检测PD大鼠的神经行为学特点 ,一管法逆转录聚合酶链反应 (RT PCR)半定量检测其中脑THmRNA的表达。 结果 造模 2 8只 ,经阿朴吗啡诱发旋转试验测试成功 2 1只 ,术后第 2、第 10周 2次旋转试验的 30min旋转次数分别为2 14 6± 30 4、2 17 5± 2 6 5 ,两者差异无显著性 (P >0 0 5 )。前肢功能试验 ,PD大鼠患肢摄食颗粒数(17 71± 4 82 )比对照大鼠 (34 43± 3 5 5 )明显减少 (P <0 0 1) ;5周后第 2次测试 ,PD大鼠患肢的摄食颗粒数 (18 86± 3 89)仍明显减少 ,与对照组 (35 86± 3 78)比较差异有显著性 (P <0 0 1) ,与其第 1次测试比较没有明显改善 (P >0 0 5 )。PD大鼠中脑的THmRNA表达水平明显降低 (P <0 0 1) ,模型组THmRNA表达约为对照组的 30 44 %。 结论 帕金森病大鼠神经行为学异常表现稳定 ,中脑内THmRNA的表达下降。
Objective To investigate the neurobehavioral characteristics and the change of tyrosine hydroxylase (TH) mRNA expression in Parkinson’s disease (PD) rats. Methods Apomorphine-induced rotation test and forelimb function test were used to detect the neurobehavioral characteristics of PD rats. Semi-quantitative RT-PCR was used to detect the expression of TH mRNA in the midbrain of PD rats. RESULTS: A total of 28 models were made and 21 successfully tested by apomorphine-induced rotation test. The number of rotations of 30 min after 2 and 10 weeks of operation was 2 14 6 ± 30 4 and 2 17 5 ± 2 There was no significant difference between the two groups (P> 0.05). In the forelimb function test, the number of pellets in the diseased limbs of PD rats (17 71 ± 4 82) was significantly lower than that of the control rats (34 43 ± 3 5 5) (P 0 01). After 5 weeks the second test PD The number of pellets in rats (18 86 ± 3 89) was still significantly decreased compared with the control group (35 86 ± 3 78) (P 0 01), but no significant difference compared with the first test Improvement (P> 0.05). The expression of TH mRNA in the midbrain of PD rats was significantly lower than that of the control rats (P <0.01). The TH mRNA expression in the model rats was about 30 44% of that in the control rats. Conclusion The neurobehavioral abnormalities in Parkinson’s disease rats are stable, and the expression of THmRNA in the midbrain decreases.
