目的:观察葛根素对糖尿病大鼠肾脏PDGF-BB表达的影响。方法:将SD大鼠随机分为正常对照组、糖尿病模型组、3种不同剂量葛根素治疗组。糖尿病大鼠模型建立后,葛根素治疗组分别给予葛根素注射液40mg/kg.d、80mg/kg.d、160mg/kg.d腹腔注射,正常对照组、糖尿病模型组腹腔注射相应量的生理盐水,实验第8周末检测空腹血糖(FBG)、尿白蛋白排泄率(UAER)、肌酐(sCr)、尿素氮(BUN);光镜和电镜观察肾皮质形态学改变;免疫组化法和Western blot法检测PDGF-BB定位和半定量表达。结果:与糖尿病模型组比较,3种不同剂量葛根素治疗组FBG、UAER、sCr、BUN明显降低,光镜及电镜下肾脏病理改变有较大改善,肾组织PDGF-BB表达减少。结论:葛根素对糖尿病大鼠肾脏有保护作用,可能与下调肾组织PDGF-BB表达有关。
Objective: To observe the effects of puerarin on the expression of PDGF-BB in the kidney of diabetic rats. Methods: SD rats were randomly divided into normal control group, diabetic model group and 3 different doses of puerarin treatment group. After the establishment of diabetic rat model, puerarin treatment group were given puerarin injection 40mg / kg.d, 80mg / kg.d, 160mg / kg.d intraperitoneal injection, normal control group, diabetic model group by intraperitoneal injection of the corresponding amount of physiology Saline, fasting blood glucose (FBG), urinary albumin excretion rate (UAER), creatinine (Cr) and blood urea nitrogen (BUN) were measured at the end of the 8th week of experiment. Morphological changes of renal cortex were observed under light microscope and electron microscope. blot assay PDGF-BB localization and semi-quantitative expression. Results: Compared with diabetic model group, FBG, UAER, sCr and BUN in three different doses of puerarin treatment group were significantly reduced, the pathological changes of kidney were improved greatly under light and electron microscope, and the expression of PDGF-BB in renal tissue was decreased. Conclusion: Puerarin has a protective effect on the kidney of diabetic rats, which may be related to the down-regulation of PDGF-BB expression in kidney.
