采用Phenomenex Luna C18柱(4.6mm×250mm,5μm),KH2PO4(0.02mol/L,pH 5.0±0.1)-乙腈(96∶4,V/V)为流动相,对乙酰氨基酚为内标,检测波长254nm,流速1.0mL/min,柱温30℃,高效液相色谱法(HPLC)测定自制阿莫西林胃粘附制剂在SD大鼠胃内的组织残留量。结果表明,阿莫西林浓度在0.52—31.38μg/mL的范围内与阿莫西林峰面积和对乙酰氨基酚峰面积之比线性关系良好(r=0.9996),检出限为0.2μg/mL,平均回收率在98.10%—101.19%之间;日内、日间精密度小于5%,测定方法稳定、可靠,专属性强,适用于生物组织样品中阿莫西林的测定。
The mobile phase consisted of Phenomenex Luna C18 column (4.6 mm × 250 mm, 5 μm), KH2PO4 (0.02 mol / L, pH 5.0 ± 0.1) -acetonitrile (96: 4, V / V) and acetaminophen as internal standard. Wavelength of 254nm, flow rate of 1.0mL / min, column temperature of 30 ℃, high performance liquid chromatography (HPLC) determination of homemade amoxicillin gastric adhesive formulations in SD rat stomach tissue residues. The results showed that the linear range of amoxicillin peak area and paracetamol peak area was 0.52-31.38μg / mL (r = 0.9996), the detection limit was 0.2μg / mL, The average recoveries ranged from 98.10% to 101.19%. The precision was less than 5% during the day and the day. The assay was stable, reliable and specific. It was suitable for the determination of amoxicillin in biological samples.
