百合鳞叶细胞培养在附加1~2mg/L6-BA的MS培养基上进行器官型态发生,在附加2mg/L2,4-D的MS培养基上进行器官发生型形态发生.电镜观察发现:两种不同途径中胚性细胞的超微结构有明显差异.器官型中胚性细胞形态相似,排列整齐规则,核大,叶绿体结构完整,线粒体与淀粉粒在胞质中分布有规律,但在器官发生型中胚性细胞之间形态有差异,排列不整齐,细胞器在胞质中无明显分布规律,但在数量上除淀粉粒外要比器官型丰富.根据观察结果,在细胞学上提出了胚性细胞中细胞器的结构分布预定其形态发生途径的新观点,即结构位置预定假说.
Lily cells were cultured on MS medium supplemented with 1 ~ 2 mg / L 6-BA and organogenesis was performed on MS medium supplemented with 2 mg / L 2,4-D. Electron microscopy showed that there were significant differences in the ultrastructure of embryogenic cells in two different pathways. Organotypic mesodermal cells were similar in shape and arranged in a regular pattern with large nuclei and chloroplast structure. The distribution of mitochondria and starch granules was regular in the cytoplasm. However, there were differences in the morphology of embryogenic cells in the organogenesis and irregular arrangement , Organelles in the cytoplasm no significant distribution, but in addition to the number of starch than the organ-type rich. Based on the observations, a new idea that the structural distribution of organelles in embryogenic cells prescribes their morphogenesis pathways has been proposed in cytology, namely, the hypothesis of the structure location hypothesis.
