目的:以对乙酰氨基酚、四氯化碳、氨基半乳糖为肝毒剂,建立急性肝损伤动物模型。方法:分别测定不同剂量对乙酰氨基酚、四氯化碳、氨基半乳糖所致急性肝损伤模型小鼠血清丙氨酸氨基转移酶(ALT)水平。结果:剂量为100 mg·kg~(-1)的对乙酰氨基酚、浓度为0.05%~1.0%四氯化碳及剂量600,800 mg·kg~(-1)的氨基半乳糖造模后均能明显升高小鼠血清ALT水平。结论:对乙酰氨基酚、四氯化碳、氨基半乳糖造模可用于保肝药的筛选与评价。
OBJECTIVE: To establish acute liver injury animal model with acetaminophen, carbon tetrachloride and galactosamine as hepatotoxic agents. Methods: Serum levels of alanine aminotransferase (ALT) in mice with acute hepatic injury induced by acetaminophen, carbon tetrachloride and galactosamine were determined at different doses. Results: Paracetamol at a dose of 100 mg · kg -1, CTC at a concentration of 0.05% -1.0%, and galactosamine at a dose of 600,800 mg · kg -1 Significantly increased serum ALT levels in mice. Conclusion: Acetaminophen, carbon tetrachloride and galactosamine can be used in the screening and evaluation of hepatoprotective drugs.
