目的:研究mTOR特异性抑制剂RAD001对胃癌SGC7901细胞增殖的影响,并探讨survivin在其中的作用。方法:常规培养胃癌SGC7901细胞,RAD001单独或联合顺铂干预,MTT法测定RAD001对胃癌SGC7901细胞增殖的影响,流式细胞术检测细胞凋亡率,免疫细胞化学和Western-blot检测药物作用后细胞中survivin蛋白的表达。结果:RAD001单独或联合顺铂干预后,①SGC7901细胞增殖受到抑制,抑制效应呈剂量依赖性并随时间的延长逐渐升高;②高浓度RAD001可以促进SGC7901细胞的凋亡;③SGC7901细胞中survivin蛋白表达下降。结论:RAD001可以通过下调细胞中survivin蛋白的表达抑制胃癌SGC7901细胞增殖,促进其凋亡。
Objective: To investigate the effect of RAD001, a specific inhibitor of mTOR, on the proliferation of gastric cancer SGC7901 cells and to explore the role of survivin in it. Methods: The gastric cancer SGC7901 cells, RAD001 alone or combined with cisplatin intervention, MTT assay RAD001 gastric cancer SGC7901 cells proliferation, apoptosis rate was detected by flow cytometry, immunocytochemistry and Western-blot detection of drug-treated cells In survivin protein expression. RESULTS: After RAD001 was administered alone or in combination with cisplatin, ① the proliferation of SGC7901 cells was inhibited and the inhibitory effect was dose-dependent and gradually increased with the prolongation of time; ② The high concentration of RAD001 promoted the apoptosis of SGC7901 cells; ③The expression of survivin protein in SGC7901 cells decline. Conclusion: RAD001 can inhibit the proliferation and promote the apoptosis of gastric cancer SGC7901 cells by down-regulating the expression of survivin protein.
