癌基因异常已知有点突变、基因过度表达、扩增及重排等。属受体型酪氨酸激酶的c-erb B2的过度表达和基因扩增与癌的恶性程度有关,故可作为癌症预后的指标。作者对9例胃癌和30例结肠癌病例进行了c-erb B2癌基因的扩增、重排及点突变分析。对异常部位,基因扩增与蛋白的过度表达、细胞外区异常与跨膜区异常的相关性进行了探讨。 结果与讨论:作者证实了Erb B2蛋白过度表达,其原因可能因基因扩增使5’端启动子结构导演而致表达调控异常。Erb B2蛋白类跨膜型受体型蛋
Abnormal oncogenes are known to have some mutations, gene overexpression, amplification and rearrangement. The overexpression and gene amplification of c-erb B2, which is a receptor type tyrosine kinase, is associated with the degree of malignancy of cancer, and thus can be used as an indicator of cancer prognosis. The authors performed c-erb B2 oncogene amplification, rearrangement, and point mutation analysis in 9 cases of gastric cancer and 30 cases of colon cancer. The relationship between abnormal regions, gene amplification and protein overexpression, abnormalities in extracellular regions, and abnormalities in transmembrane regions was discussed.
