目的 :研究重组人白细胞介素 13 (rhIL 13 )对巨核细胞系 -Dami细胞原癌基因c mpl表达的影响。方法 :实验组细胞培养系统中分别加入 2 5ng/ml,5 0ng/ml,10 0ng/mlrhIL 13 ,对照组不加任何细胞因子。用RT PCR法检测c mplmRNA的表达。结果 :经 2 5ng/ml、5 0ng/ml、10 0ng/mlrhIL 13处理过的实验组Dami细胞较之对照组其c mplmRNA的表达分别提高 2 4 .8%、2 7.9%和 3 1.5 %。结论 :rhIL 13增强了Dami细胞原癌基因c mpl表达 ;rhIL 13促进Dami细胞增殖的部分机制可能是通过上调c mpl的表达来实现的。
Objective: To investigate the effect of recombinant human interleukin-13 (rhIL-13) on cmpl expression in megakaryocytic line-Dami cells. Methods: The experimental group cell culture system were added 25ng / ml, 50ng / ml, 10ng / mlrhIL13, the control group without any cytokines. The expression of c mpl mRNA was detected by RT PCR. Results: Compared with the control group, the Dami cells treated with 25ng / ml, 50ng / ml and 10ng / ml rhIL13 showed an increase of 24.8%, 29.7% and 31.5% respectively compared with the control group. CONCLUSION: rhIL 13 enhances the expression of proto-oncogene cmpl in Dami cells. Part of the mechanism by which rhIL 13 promotes proliferation of Dami cells may be through the up-regulation of cmpl expression.
