采用单因子和正交实验优化灵芝子实体粗多糖微波法提取工艺和硫酸蒽酮比色法,建立了灵芝子实体粗多糖微波提取和检测方法。过60目筛的粉末作为样品,预浸1.5 h,水提液pH 8.0,料水比1∶70,微波功率400 W,微波时间20 min,提取2次,然后采用90%硫酸-蒽酮法,水浴10 min进行多糖比色检测。通过多糖清除DPPH自由基的实验,初步探讨了微波法提取灵芝多糖的抗氧化能力。
The single factor and orthogonal experiments were used to optimize the crude polysaccharide extraction method and the anthrone sulfuric acid colorimetric method. The microwave extraction and detection methods of crude polysaccharides from Ganoderma lucidum were established. 60 mesh sieve powder as sample, pre-immersion 1.5 h, water extract pH 8.0, material to water ratio 1:70, microwave power 400 W, microwave time 20 min, extracted twice, then 90% sulfuric acid - , Water bath for 10 min polysaccharide colorimetric assay. Through the polysaccharide scavenging DPPH free radical experiments, the antimicrobial activity of Ganoderma lucidum polysaccharides extracted by microwave was preliminarily discussed.
