目的:探讨HCV核心蛋白对肝细胞侵袭性的影响。方法:构建HCV核心蛋白真核表达载体pIRES-core,稳定转染L02细胞,通过间接免疫荧光和Western印迹方法检测核心蛋白的表达,电镜观察肝细胞的形态变化,用体外迁移侵袭力实验测定转染HCV核心蛋白肝细胞的迁移侵袭的能力。结果:未转染细胞L02和转染空载体细胞相比,转染HCV核心蛋白细胞表达基质金属蛋白酶诱导物(EMMPRIN)的水平升高,并且该细胞的迁移侵袭能力明显升高(P<0.01)。结论:HCV核心蛋白可能通过上调EMMPRIN的表达以提高肝细胞的迁移侵袭能力,为进一步探讨核心蛋白如何诱导肝细胞侵袭性奠定了基础。
Objective: To investigate the influence of HCV core protein on the invasiveness of hepatocytes. Methods: The eukaryotic expression vector pIRES-core of HCV core protein was constructed and stably transfected into L02 cells. The expression of core protein was detected by indirect immunofluorescence and Western blotting. The morphological changes of hepatocytes were observed by electron microscopy. The ability to infect the migration and invasion of HCV core protein hepatocytes. Results: The expression of EMMPRIN in transfected HCV core cells was higher than that in transfected empty vector cells (P <0.01) ). Conclusion: HCV core protein may enhance the invasion and migration of hepatocytes by up-regulating the expression of EMMPRIN, which lays the foundation for further exploring how the core protein induces hepatocellular invasion.
