Objective To investigate the role of complete Freund’s adjuvant (CFA) in preventing insulitis and diabetes mellitus in non-obese diabetic (NOD) mice. Methods 3-week-old NOD female rats were randomly divided into CFA group and PBS control group. The CFA group and the PBS group were given a single injection of 50μl of CFA and 50μl of PBS at the 6th and 12th week At 30 weeks of age, pancreatic β-cell apoptosis, insulitis and diabetes were observed. Results At 6 and 12 weeks, the scores of insulitis and β-cell apoptosis in CFA group were significantly lower than those in PBS group (P <0.05, P <0.01), and the scores of insulitis at 6 weeks and 12 weeks β-cell apoptosis rate is positively correlated. At 30 weeks of age, only 1 out of 11 mice in the CFA group developed diabetes mellitus (1/11), significantly less than in the PBS group [9 of 11 (9/11)], P = 0 0 01. Conclusion Early administration of CFA NOD mice can significantly reduce the occurrence of insulitis and diabetes mellitus, its mechanism and inhibition of β-cell apoptosis.
