目的探讨多位点序列分型方法在沙门菌分型鉴定研究中的应用,建立水产品中沙门菌监测的分子分型数据库。方法选取沙门菌的7对管家基因aro C、dna N、hem D、his D、pur E、suc A和thr A作为靶标基因,对分离鉴定的47株沙门菌样本进行PCR扩增、测序及序列分析。结果 47株沙门菌分离株有20个ST型,其中ST11为优势ST型,占分离株的27.66%。此外,本研究新鉴定出3个ST型,分别为ST1808、ST1809和ST1810。结论多位点序列分型为水产源性沙门菌的分类研究提供了信息,对研究沙门菌的进化关系、鉴定溯源及预防控制预警提供了技术支持和数据参考。
Objective To investigate the application of multilocus sequence typing in Salmonella typing and to establish a molecular typing database for Salmonella surveillance in aquatic products. Methods Seven pairs of housekeeping genes aro C, dna N, hem D, his D, pur E, suc A and thr A of Salmonella were selected as target genes, 47 strains of Salmonella isolated and identified were amplified by PCR, sequenced and sequenced analysis. Results Totally 47 Strains of Salmonella isolated from ST type, of which ST11 was predominant ST type, accounting for 27.66% of the isolates. In addition, three ST types were newly identified in this study, namely ST1808, ST1809 and ST1810. Conclusions Multi-site sequence typing provided information for taxonomic study of Salmonella typhi, and provided technical support and data reference for studying the evolutionary relationship, identification and tracing of SARS and prevention and control of Salmonella.
