CTL 可能存在着多种溶解靶细胞的机制。虽然近年来应用T 细胞克隆对CTL 溶解机制进行了很多研究,但对新鲜分离的或培养的人外周血T 细胞溶解机制了解甚少。先用Percoll 分离人PBL 中高密度T 细胞,然后用流式细胞仪纯化CD4~+或CD8~+T 细胞亚群。用IL-2和IL-2/OKT_3单抗分别激活CD8~+和CD4~+T 细胞,通过Northern 印迹法检测其mRNA 表达,并且在4小时和18小时~(51)Cr 释放试验中,检测两种T 细胞亚群对NP(萘酚)修饰的TNF 敏感靶细胞L959
CTL may exist a variety of mechanisms to dissolve target cells. Although many studies have been made on the mechanism of CTL lysis using T cell clones in recent years, little is known about the mechanism of T cell lysis in freshly isolated or cultured human peripheral blood.
