报道两种长度的HCV核心基因片段在E.coli中的表达结果。从HCV感染者的血清扩增出核心区基因,利用表达质粒pQE-30,构建了两种重组质粒,其插入片段分别为HCV的全长核心区基因(c573)和“截断型”基因(c375)。经IPTG诱导,含两种质粒的菌株都表达了目的蛋白,c573表达的蛋白分子量为22kD,表达量占菌体蛋白的8.7%;c375表达的蛋白分子量为19kD,表达量占菌体蛋白的39.9%。经Ni-NTA金属螯合层析纯化,获得了高纯度的目的蛋白。这两种蛋白均可特异地与丙型肝炎病人血清发生反应,表明具有良好的抗原活性
Two lengths of HCV core gene fragment are reported in E. coli. Expression in E. coli. The core region genes were amplified from the sera of HCV-infected patients. Two recombinant plasmids were constructed by using the expression plasmid pQE-30. The inserts contained the full-length core region gene (c573) and the truncated gene (c375 ). After IPTG induction, the two plasmids expressed the target protein. The protein expressed by c573 had a molecular weight of 22kD and accounted for 8.7% of the total bacterial protein. The protein expressed by c375 was 19kD, Of the 39.9%. Purified by Ni-NTA metal chelate chromatography to obtain a high purity of the target protein. Both of these proteins can specifically react with hepatitis C patient serum, indicating that they have good antigenic activity
