目的对比观察滋阴活血方药(nourishing yin and promoting blood-flow recipe,NYPBR)和尿酸盐(urate)对糖尿病大鼠牙周组织结构的影响。方法选用6月龄雄性Wistar大鼠38只随机分为4组,对照组8只、糖尿病组、中药组和西药组各10只。建立大鼠糖尿病模型,中药组给予NYPBR,西药组给予urate。给药后12周处死大鼠,取上颌骨标本,行组织学及骨计量学观察。结果①组织学观察:对照组牙周组织无明显变化;糖尿病组大鼠12周时牙龈上皮和固有层中度炎细胞浸润,牙龈和牙周膜内毛细血管数目增多、扩张充血,纤维排列紊乱和断裂,骨吸收陷窝多,陷窝内见破骨细胞,牙槽嵴顶无吸收;中药组、西药组牙周组织结构无明显异常,结构与对照组相似。②骨计量学观察:12周时大鼠牙周结缔组织附着丧失量和牙槽骨嵴高度丧失量各组间无差异(P>0.05)。对照组、中药组、西药组成骨细胞数和破骨细胞数无明显差异(P>0.05),成骨细胞数明显高于糖尿病组(P<0.05),破骨细胞数明显低于糖尿病组(P<0.05)。结论 NYPBR和urate可有效预防糖尿病造成的牙周组织受损。
Objective To observe the effects of nourishing yin and promoting blood-flow recipe (NYPBR) and urate on the periodontal structure in diabetic rats. Methods 38 male Wistar rats aged 6 months were randomly divided into 4 groups, 8 in the control group, 10 in the diabetes group, the traditional Chinese medicine group and the western medicine group. The rat model of diabetes was established. NYPBR was given to the traditional Chinese medicine group and urate was given to the western medicine group. The rats were sacrificed 12 weeks after the administration, and the maxillary specimens were taken for histological observation and bone metrology. Results ① Histological observation showed no significant changes in the periodontal tissues in the control group. In the diabetic group, the infiltration of inflammatory cells in the gingival epithelium and lamina propria at 12 weeks increased the number of capillaries in the gingival and periodontal ligament, dilated and hyperemic, And fracture, the majority of bone resorption lacunae, see the osteoclasts within the nest, alveolar crest no absorption; Chinese medicine group, Western medicine group no significant abnormalities in the periodontal tissue structure similar to the control group. ② Bone metrology observation: There was no significant difference in the loss of connective tissue attachment and alveolar ridge height between the two groups at 12 weeks (P> 0.05). The numbers of osteocytes and osteoclasts in the control group, the traditional Chinese medicine group and the western medicine group were not significantly different (P> 0.05), the number of osteoblasts was significantly higher than that in the diabetic group (P <0.05), the number of osteoclasts was significantly lower than that in the diabetic group P <0.05). Conclusion NYPBR and urate can effectively prevent periodontal tissue damage caused by diabetes.
