将苦参碱给小鼠肌肉注射,其 LD_(50)为74.15±6.14mg/kg。苦参碱与氢化可的松相似,肌肉注射都能明显对抗巴豆油诱发小鼠和大鼠耳壳的炎症,长期给药时其作用随剂量增加而增强。它对大鼠后肢由角叉菜胶诱发的炎症和对小鼠腹腔注射冰醋酸诱发的渗出性炎症均有明显的抑制作用;但对大鼠由埋藏棉球诱发肉芽组织增生的慢性炎症却无影响,而氢化可的松则有明显的对抗作用。在分析苦参碱的抗炎症作用机理时发现,它对垂体—肾上腺系统无明显影响。管内实验发现苦参碱能明显对抗血清蛋白加热变性的作用;对红细胞的溶血现象也有抑制作用。
Matrine was intramuscularly injected to mice with an LD_(50) of 74.15±6.14 mg/kg. Matrine is similar to hydrocortisone in that intramuscular injection can significantly counteract the inflammation of the ear shells of mice and rats induced by croton oil, and its effects are enhanced with increasing doses during long-term administration. It has a significant inhibitory effect on carrageenan-induced inflammation in the hind limbs of rats and exudative inflammation induced by intraperitoneal injection of glacial acetic acid in rats; however, it induces chronic inflammation of granulation tissue in rats induced by buried cotton balls. No effect, while hydrocortisone has obvious antagonistic effects. In analyzing the anti-inflammatory mechanism of matrine, it was found that it had no significant effect on the pituitary-adrenal system. In-tube experiments showed that matrine can significantly counteract the heat-denaturation effect of serum proteins and also inhibit the hemolysis of red blood cells.
