在低氧条件下,猪肺动脉内皮细胞(PAEC)对猪肺动脉平滑肌细胞(PASMC)增殖调控,是研究低氧性肺动脉高压时肺动脉结构重组的重要内容。应用MTT比色法、DNA荧光测定法,观察了低氧PAEC条件培养基(HECCM)中多胺促PASMC增殖的作用。结果表明,在PAEC培养液中加入鸟氨酸脱梭酶抑制剂二氟甲基鸟氨酸(DFMO),可阻止HECCM促PASMC增殖的效应。故认为低氧刺激PAEC产生的多胺亦是HECCM中促PASMC增殖的一个重要因子。
Under hypoxic conditions, the regulation of proliferation of porcine pulmonary artery smooth muscle cells (PASMCs) by porcine pulmonary artery endothelial cells (PAECs) is an important part of pulmonary structural remodeling in hypoxic pulmonary hypertension. The effect of polyamine on the proliferation of PASMC in hypoxic PAEC conditioned medium (HECCM) was observed by MTT assay and DNA fluorescence assay. The results showed that addition of ornithine deltorubicin inhibitor difluoromethylornithine (DFMO) in PAEC medium prevented the HECCM from promoting the proliferation of PASMC. Therefore, hypoxia-induced PAEC production of polyamines is also HECCM in promoting PASMC proliferation is an important factor.
