目的:应用PCR-焦磷酸测序技术对食物中毒标本中沙门菌进行快速检测。方法:疑似沙门菌食物中毒的相关标本9份。前增菌后,进行核酸提取及PCR-焦磷酸测序技术检测。同时按GB/T4789.4-2008国标法进行细菌分离培养,利用VITEK全自动微生物鉴定仪对可疑菌进行生化反应检测,血清学试验鉴定血清型。结果:PCR-焦磷酸测序结果显示9份标本中3份沙门菌阳性。用分离鉴定方法也仅在该3份标本中分离出沙门菌。结论:PCR-焦磷酸测序技术可用于沙门菌食物中毒的快速检测。
Objective: To detect Salmonella in food poisoning samples by PCR-pyrosequencing. Methods: Nine samples were suspected of Salmonella food poisoning. After pre-enrichment, nucleic acid extraction and PCR-pyrosequencing were performed. At the same time according to GB / T4789.4-2008 GB method of bacterial isolation and culture, the use of VITEK automatic microbial identification of suspicious bacteria for biochemical reactions, serological tests to identify serotypes. Results: PCR-pyrosequencing revealed 3 Salmonella positive in 9 samples. Salmonella was isolated only from the three specimens by isolation and identification method. Conclusion: PCR-Pyrosequencing can be used for the rapid detection of Salmonella food poisoning.
