口虾蛄乙酸乙酯提取物对鼻咽癌CNE-2Z细胞增殖和周期的影响

目的:观察口虾蛄乙酸乙酯提取物对鼻咽癌CNE-2Z细胞增殖和周期的影响。方法:MTT比色法检测药物对细胞的增殖抑制率,[3H]-TdR标记法测定药物对细胞DNA合成的影响,流式细胞术分析细胞周期。结果:口虾蛄乙酸乙酯提取物对CNE-2Z细胞的增殖和DNA合成有显著的抑制作用,并呈明显的时间-剂量依赖关系,当提取物浓度>400μg/mL时,其对CNE-2Z细胞的掺入抑制率高于浓度为20μg/mL的5-FU对该细胞系的掺入抑制率。流式细胞术检测结果显示,随着乙酸乙酯提取物浓度的增加,G1期和G2/M期细胞所占的百分比逐渐降低,S期细胞所占百分比逐渐增加,P<0.01。结论:口虾蛄乙酸乙酯提取物能抑制CNE-2Z细胞的增殖效应,其机制可能与引起细胞周期阻滞于S期有关。 OBJECTIVE: To observe the effects of ethyl acetate extract of Eupatorium mulatta on the proliferation and cell cycle of nasopharyngeal carcinoma CNE-2Z cells. Methods: MTT colorimetric assay was used to detect the inhibitory effect of drugs on cell proliferation. The effect of drugs on the DNA synthesis was assayed by [3H] -TdR labeling. Cell cycle was analyzed by flow cytometry. Results: Ethyl acetate extract of Eupatorium japonicus significantly inhibited the proliferation and DNA synthesis of CNE-2Z cells in a time-and dose-dependent manner. When the extract concentration was> 400 μg / mL, the CNE- The incorporation inhibition rate of 2Z cells was higher than that of 5-FU at a concentration of 20 μg / mL. Flow cytometry results showed that with the increase of ethyl acetate extract concentration, the percentage of cells in G1 phase and G2 / M phase decreased gradually, and the percentage of S phase cells increased gradually (P <0.01). CONCLUSION: Ethyl acetate extract can inhibit the proliferation of CNE-2Z cells. The mechanism may be related to cell cycle arrest in S phase.
