目的观察莫诺苷对局灶性脑缺血再灌注大鼠皮层血管内皮生长因子(VEGF)和成纤维生长因子-2(FGF-2)表达的影响。方法 25只成年雄性Sprague-Dawley大鼠随机分为假手术组、模型组、莫诺苷小剂量组(30 mg/kg)、莫诺苷中剂量组(90 mg/kg)和莫诺苷大剂量组(270 mg/kg)。采用改良线栓法制备大鼠大脑中动脉栓塞(MCAO)30 min再灌注模型。再灌注7 d后利用蛋白免疫印迹法检测梗死侧皮层VEGF和FGF-2蛋白表达。结果与假手术组相比,缺血再灌注7 d后,各组梗死侧大脑皮层VEGF、FGF-2的表达均有增加(P<0.05);莫诺苷小、中、大剂量组VEGF表达进一步增加(P<0.05);莫诺苷大剂量组FGF-2的表达进一步显著增加(P<0.001)。结论莫诺苷可以促进局灶性脑缺血再灌注后VEGF和FGF-2的表达,可能有助于促进缺血性脑损伤后的血管新生。
Objective To observe the effects of morroniside on the expression of vascular endothelial growth factor (VEGF) and fibroblast growth factor-2 (FGF-2) in focal cerebral ischemia-reperfusion rats. Methods Twenty-five male Sprague-Dawley rats were randomly divided into sham operation group, model group, low dosage of morroniside (30 mg / kg), middle dosage of morroniside (90 mg / kg) Dose group (270 mg / kg). A rat model of middle cerebral artery occlusion (MCAO) 30 min reperfusion was established by modified suture method. The expression of VEGF and FGF-2 in the infarct cortex was detected by Western blot 7 days after reperfusion. Results Compared with the sham operation group, the expression of VEGF and FGF-2 in the infarcted side of each group increased after 7 d of ischemia / reperfusion (P <0.05). The expression of VEGF in small, middle and large dose of morroniside (P <0.05). The expression of FGF-2 in the high-dose morroniside group was significantly increased (P <0.001). Conclusion Morroniside can promote the expression of VEGF and FGF-2 after focal cerebral ischemia and reperfusion, which may be helpful to promote angiogenesis after ischemic brain injury.
