[目的]探讨选择性环氧化酶-2(COX-2)抑制剂celecoxib对人卵巢癌耐药细胞株SKOV-3/DDP的增殖抑制作用并初步探讨其机制。[方法]用不同浓度celecoxib作用SKOV-3/DDP细胞不同时间后,MTT法检测细胞增殖抑制情况,RT-PCR法检测细胞中survivin及GST-πmRNA的表达情况,FCM法检测细胞survivin及GST-π蛋白表达情况。[结果]celecoxib可以抑制SKOV-3/DDP细胞的增殖,呈药物浓度及时间依赖性。RT-PCR法检测发现与未处理组相比,celecoxib可下调SKOV-3/DDP细胞中survivin及GST-πmRNA的表达(P<0.01),FCM法检测显示celecoxib可下调细胞中survivin及GST-π蛋白表达(P<0.01)。[结论]celecoxib能抑制SKOV-3/DDP细胞增殖,并呈浓度及时间依赖性,其机制可能与下调survivin及GST-π表达相关。
[Objective] To investigate the inhibitory effect of celecoxib, a selective cyclooxygenase-2 (COX-2) inhibitor, on the proliferation of human ovarian cancer cell line SKOV-3 / DDP and its possible mechanism. [Methods] After treated with different concentrations of celecoxib for different time periods, MTT assay was used to detect the proliferation of SKOV-3 / DDP cells. The expression of survivin and GST-πmRNA was detected by RT-PCR. The expressions of survivin and GST- π protein expression. [Results] celecoxib inhibited the proliferation of SKOV-3 / DDP cells in a concentration and time-dependent manner. The results of RT-PCR showed that celecoxib downregulated the expression of survivin and GST-πmRNA (P <0.01) in SKOV-3 / DDP cells compared with untreated cells. The expression of survivin and GST-π in celecoxib down- Protein expression (P <0.01). [Conclusion] celecoxib can inhibit the proliferation of SKOV-3 / DDP cells in a concentration-and time-dependent manner, which may be related to the down-regulation of survivin and GST-π expression.
