目的:研究三氧化二砷体外能否诱导宫颈癌细胞株(H eLa细胞)发生凋亡及其相关机制。方法:采用透射电镜、D N A Ladder、TU N E L的方法进行凋亡检测,用E C L W esternblot的方法检测凋亡过程中相关基因表达的变化。结果:三氧化二砷体外作用于H eLa细胞后,透射电镜观察到了凋亡小体;D N A Ladder法检测到了凋亡时D N A降解形成的梯带;TU N E L法也检测到H eLa细胞有D N A的断裂。在凋亡过程中,凋亡相关基因bcl-2的表达下调,而p53基因的表达没有明显改变。结论:三氧化二砷体外能诱导H eLa细胞发生凋亡,其机制与bcl-2基因的表达下调有关。
Objective: To investigate whether arsenic trioxide can induce the apoptosis of cervical cancer cell line H eLa in vitro and its related mechanism. Methods: Apoptosis was detected by transmission electron microscopy, D N A Ladder and TU N E L, and the changes of related gene expression during apoptosis were detected by using the E C L W esternblot method. RESULTS: Arsenic trioxide (H 2 O 3) was applied to H eLa cells in vitro and apoptotic bodies were observed by transmission electron microscopy. Ladder of D N A degradation was detected by D N A Ladder method and D N A was also detected by TU N E L method. In the process of apoptosis, the expression of bcl-2 was down-regulated while the expression of p53 was not significantly changed. CONCLUSION: Arsenic trioxide can induce apoptosis in H eLa cells in vitro, and its mechanism is related to the down-regulation of bcl-2 gene expression.
